Biological Background: HSP27/HSPB1 Function and Localization

  • Also known as 28 kDa heat shock protein, Estrogen-regulated 24 kDa protein, and Stress-responsive protein 27 (SRP27).
  • Small heat shock protein that functions as a molecular chaperone, maintaining denatured proteins in a folding-competent state and playing a role in stress resistance and actin organization. Related references: PMID:10383393, PMID:20178975, PMID:19166925
  • Through its chaperone activity, may regulate phosphorylation and axonal transport of neurofilament proteins. Related references: PMID:23728742
  • Localizes to the cytoplasm, nucleus, and cytoskeleton, including spindle structures.
  • Detected in all tissues, with highest expression in heart and striated/smooth muscle.
  • Phosphorylation at Ser82 (and other residues) modulates HSPB1 chaperone function and client protein interactions.
  • Mutations in HSPB1 are linked to Charcot-Marie-Tooth disease type 2F (CMT2F) and distal hereditary motor neuropathy (dHMN).

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic phosphopeptide; always include phosphatase inhibitors in sample preparation to preserve phosphorylation state.
  • For WB validation, treat lysates with lambda phosphatase—loss of signal confirms phospho-specificity.
  • Optimize antibody dilution for your specific sample system; consider a titration series to determine the ideal working concentration.
  • For ELISA, a phosphorylated peptide coating antigen is recommended to ensure phospho-specific detection.
