Biological Background: HDAC5 Function and Localization
- HDAC5 (Histone deacetylase 5, antigen NY-CO-9) is a class II histone deacetylase that catalyzes the removal of acetyl groups from lysine residues on core histones H2A, H2B, H3, and H4. Related references: PMID:10220385
- Functions as a transcriptional repressor by promoting chromatin compaction, playing roles in cell cycle progression, development, and epigenetic silencing.
- Acts as a corepressor for transcription factors, including MEF2C during muscle differentiation; its nuclear-cytoplasmic shuttling regulates myocyte gene expression.
- In association with RARA, mediates deacetylation and repression of microRNA-10a, modulating inflammatory responses. Related references: PMID:28167758
- Involved in MTA1-mediated epigenetic regulation of ESR1 expression in breast cancer.
- Localizes predominantly in the nucleus but can shuttle to the cytoplasm in a phosphorylation-dependent manner. Keywords: Nucleus, Cytoplasm.
- Widely expressed across tissues, including heart, brain, kidney, pancreas, liver, skeletal muscle, and placenta. Related references: PMID:10220385
- The protein has a molecular weight of ~122 kDa and is subject to multiple post-translational modifications, including phosphorylation, acetylation, and ubiquitination.
Experimental Guidance and Technical Tips
- This antibody was raised against a synthetic phosphopeptide surrounding serine 498 of human HDAC5; it is expected to specifically detect phosphorylated HDAC5 at that site.
- Suitable for Western blot (WB) analysis. A band corresponding to the predicted molecular weight of ~122 kDa may be observed. Validate the antibody in your specific sample system.
- Also validated for ELISA; consider optimizing coating conditions and antibody dilutions for your assay format.
- Reported to cross-react with human, mouse, and rat HDAC5. For species not listed, perform sequence alignment of the target epitope to assess potential reactivity.
- For Western blot, use appropriate phospho-specific controls (e.g., phosphatase treatment) to confirm phospho-specificity.
