Biological Background: FOXO3A Function and Localization

  • FOXO3A is a transcriptional activator that recognizes the DNA sequence 5'-[AG]TAAA[TC]A-3' and regulates critical processes including apoptosis and autophagy.
    Related references: PMID:10102273, PMID:16751106, PMID:21329882
  • In skeletal muscle, FOXO3A acts as a positive regulator of autophagy by entering the nucleus upon dephosphorylation and activating autophagy genes (GABARAP1L, MAP1LC3B, ATG12), leading to protein breakdown.
  • The protein triggers apoptosis in the absence of survival factors, including neuronal cell death under oxidative stress.
    Related references: PMID:10102273, PMID:16751106
  • FOXO3A participates in post-transcriptional regulation of MYC by promoting expression of miR-34b and miR-34c, which bind the MYC 3'UTR and inhibit translation.
    Related references: PMID:21329882
  • Upon metabolic stress, FOXO3A translocates to mitochondria where it enhances mtDNA transcription.
    Related references: PMID:23283301
  • FOXO3A is a key regulator of regulatory T-cell (Treg) differentiation through activation of FOXP3 expression.
    Related references: PMID:30513302
  • Subcellular localization includes the cytoplasm, cytosol, nucleus, mitochondrial matrix, and mitochondrial outer membrane. Its activity is regulated by post-translational modifications such as phosphorylation, acetylation, and ubiquitination.
  • FOXO3A is ubiquitously expressed across tissues.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic phosphorylated peptide around Ser253; the antibody may preferentially recognize the phosphorylated form of FOXO3A. For Western blot, consider treating samples with phosphatase to confirm phosphorylation-specific detection.
  • For immunohistochemistry (IHC-P), antigen retrieval conditions and tissue fixation methods should be optimized. Validate staining specificity with appropriate positive and negative tissue controls.
  • The antibody cross-reacts with human, mouse, and rat FOXO3A, making it suitable for multispecies studies. Verify species-specific reactivity in your experimental system.
