Biological Background: FOXO1 Function and Localization

  • FOXO1 is a transcription factor and the main target of insulin signaling, regulating metabolic homeostasis in response to oxidative stress. Related references: PMID:10358076 PMID:12228231 PMID:15220471
  • It binds to insulin response elements (IRE) and Daf-16 family binding elements (DBE), with its activity suppressed by insulin. Related references: PMID:10358076
  • FOXO1 shuttles between the cytoplasm and nucleus, and its subcellular localization is regulated by post-translational modifications including phosphorylation at sites such as Ser319.
  • In hepatocytes, it promotes gluconeogenesis by directly activating the expression of PPARGC1A and G6PC1. Related references: PMID:17024043
  • FOXO1 is an important regulator of cell death, acting downstream of CDK1, PKB/AKT1, and STK4/MST1, and promotes neural cell death. Related references: PMID:18356527 PMID:19221179
  • It is required for autophagic cell death induction in response to starvation or oxidative stress in a transcription-independent manner. Related references: PMID:20543840
  • FOXO1 positively regulates apoptosis in cardiac smooth muscle cells and controls endothelial cell viability and apoptosis via PPIA/CYPA-dependent transcription. Related references: PMID:19483080 PMID:31063815
  • Tissue expression: abundantly expressed in skeletal muscle and ovary, with lower levels in heart, liver, and other tissues; expressed in umbilical endothelial cells at the protein level. Related references: PMID:9479491 PMID:19483080

Experimental Guidance and Technical Tips

  • This antibody was raised against a synthetic phosphorylated peptide corresponding to residues surrounding Ser319 of human FOXO1. For detection of phosphorylation at this site, consider using phosphatase inhibitors in lysis buffers to preserve the phospho-epitope.
  • In western blotting, a band consistent with the predicted molecular weight of FOXO1 (~70 kDa) may be observed. We recommend validation in your experimental system with appropriate positive and negative controls.
  • For immunohistochemistry (paraffin), antigen retrieval methods and antibody titration should be optimized for the specific tissue type and fixation conditions.
  • Given its cross-reactivity with mouse and rat, this antibody can be a useful tool for comparative studies across these species; however, sequence alignment of the immunogen region is advised to confirm epitope conservation.
