Biological Background: Ezrin/Radixin/Moesin Function and Localization

  • Ezrin (also known as cytovillin, villin-2, p81) is a protein encoded by the EZR gene in humans. Together with radixin and moesin, it constitutes the ERM protein family that links the actin cytoskeleton to the plasma membrane.
  • Ezrin is involved in connecting major cytoskeletal structures to the plasma membrane, and in epithelial cells it is required for the formation of microvilli and membrane ruffles on the apical pole.
  • Along with PLEKHG6, ezrin participates in normal macropinocytosis.
  • Subcellularly, ezrin localizes to the apical cell membrane, microvillus and ruffle membranes, cytoplasm, cell cortex, and cytoskeleton, consistent with its role in cell projection maintenance.
  • Tissue expression includes cerebral cortex, basal ganglia, hippocampus, hypophysis, optic nerve, and intestinal epithelial microvilli; it is preferentially expressed in astrocytes among brain regions.
  • This antibody recognizes the phosphorylated forms at the conserved C-terminal threonine residues (T567 in ezrin, T564 in radixin, T558 in moesin), a modification critical for ERM activation and open conformation.

  • The immunogen is a synthetic phosphopeptide corresponding to the conserved activation domain; ensure phosphorylation state is preserved by using phosphatase inhibitors during cell lysis.
  • For Western blot, expected bands include ~69 kDa (ezrin), ~68 kDa (radixin), and ~80 kDa (moesin), with the reported 23-71 kDa range possibly reflecting fragments or isoform variants; validate in relevant cell lines.
  • This antibody is applicable for ELISA; consider optimizing coating conditions and using phospho- and non-phospho peptide controls for specificity.
  • Cross-reactivity with human, mouse, and rat has been indicated; for other species, preliminary testing is recommended.
