Biological Background: EZH2 Function and Localization
- EZH2 is a histone-lysine N-methyltransferase, also known as enhancer of zeste homolog 2, ENX-1, or KMT6, and functions as the catalytic subunit of the PRC2 complex.

Experimental Guidance and Technical Tips
- The immunogen used to generate this antibody is a synthetic peptide carrying phosphorylation at Thr311; therefore, the antibody is expected to specifically recognize the phosphorylated form of EZH2 at this site. Confirm specificity by comparing signals in phosphatase-treated versus untreated samples.
- For western blot (WB), EZH2 migrates at ~85kDa. Use appropriate positive controls such as HEK293T or HeLa cells, and validate detection of the phospho-T311 form under conditions that induce EZH2 phosphorylation.
- ELISA can be employed for quantitative analysis of phospho-EZH2 (T311) levels; consider using a capture antibody in combination with this pAb for sandwich ELISA.
- Cross-reactivity with rat has been observed; thus, this antibody may be suitable for rodent model studies. Validate in the relevant tissue or cell line system to ensure specificity.
