Biological Background: Estrogen Receptor Alpha Function and Localization

  • Estrogen receptor alpha (ER-alpha), encoded by the ESR1 gene, is a steroid hormone receptor and member of the nuclear receptor subfamily 3 group A (NR3A1). It functions as a ligand-activated transcription factor, regulating gene expression critical for cell proliferation and differentiation in target tissues.
  • Upon estrogen binding, ER-alpha undergoes a conformational change that facilitates homodimerization and binding to estrogen response elements (EREs) in DNA. It also interacts with other transcription factors such as AP-1 (c-Jun/c-Fos) and Sp1 to mediate ERE-independent signaling. Coactivator complexes are recruited via LXXLL motifs.

  • ER-alpha shuttles between the nucleus, cytoplasm, and cell membrane. It is also associated with the Golgi apparatus, enabling both genomic and rapid non-genomic signaling pathways, including activation of kinase cascades.
  • The receptor is subject to multiple post-translational modifications including phosphorylation, glycosylation, palmitoylation, and ubiquitin-like conjugation. It contains zinc finger motifs and a ligand-binding domain, with a molecular weight of approximately 66 kDa.

  • For western blotting, the predicted band size is ~66 kDa. Validate detection in human cell lines known to express ER-alpha, such as MCF-7.
  • In immunofluorescence/ICC, anticipate nuclear, cytoplasmic, and membrane localization. Co-stain with compartment markers to confirm specificity.
  • As a polyclonal antibody, lot-to-lot consistency may vary; include positive and negative controls, and consider a blocking peptide for phospho-specificity verification.
