Biological Background: ERK1/2 Function and Localization

  • Mitogen-activated protein kinase 3 (MAPK3/ERK1) is a serine/threonine kinase that acts as an essential component of the MAP kinase signal transduction pathway, mediating diverse biological functions such as cell growth, adhesion, survival, and differentiation. Related references: PMID:34497368
  • ERK1 phosphorylates over 160 known substrates localized in the nucleus (e.g., transcription factors ATF2, ELK1, FOS) and cytosol (e.g., cytoskeletal elements, regulators of apoptosis and translation), enabling broad control over transcription, translation, and apoptosis.   • In resting cells, ERK1 is predominantly cytoplasmic but upon stimulation translocates to the nucleus. It also localizes to membrane caveolae, cell junctions, and focal adhesions, underscoring its roles in signal integration and cytoskeletal reorganization.

Experimental Guidance and Technical Tips

  • Based on the synthetic phosphopeptide immunogen around T202/Y204 of human ERK1, this antibody is designed to recognize the active, dual-phosphorylated form of ERK1 and cross-reacts with the corresponding phosphorylated sites (T185/Y187) in ERK2 due to high sequence homology. Include phosphatase inhibitors during sample preparation to preserve phosphorylation.
  • For Western Blot, expect a band at ~42–44 kDa. To maintain phospho-signal integrity, use mild stripping buffers or prepare fresh samples for repeated probing rather than harsh stripping.
  • In IHC-P, standard antigen retrieval (e.g., citrate buffer, pH 6.0) may be applied. Include both stimulated and unstimulated tissue controls to confirm phospho-specificity.
  • For ELISA, a sandwich format using a pan-ERK capture antibody paired with this detection antibody can quantify active ERK1/2 in lysates or homogenates.
