Biological Background: eNOS Function and Localization

  • Nitric oxide synthase 3 (eNOS) is an endothelial-specific enzyme that catalyzes the production of nitric oxide (NO), a key signaling molecule in the cardiovascular system.
  • eNOS-derived NO induces vascular smooth muscle relaxation through a cGMP-mediated signal transduction pathway, playing a critical role in blood pressure regulation. Related references: PMID:1378832
  • NO generated by eNOS also mediates VEGF-induced angiogenesis in coronary vessels and promotes platelet activation, contributing to hemostasis.
  • An alternatively spliced isoform of eNOS lacks catalytic activity and may act as a dominant-negative regulator by forming heterodimers with the active full-length enzyme.
  • eNOS localizes to the cell membrane, caveolae, cytoplasm, cytoskeleton, and Golgi apparatus; membrane targeting is facilitated by N-terminal myristoylation and palmitoylation.
  • Expression has been detected in platelets, placenta, liver, and kidney.
  • eNOS is a phosphoprotein whose activity is modulated by phosphorylation, calcium/calmodulin binding, and cofactors such as FAD, FMN, heme, iron, zinc, and NADP.

Experimental Guidance and Technical Tips

  • When performing Western blot, include phosphatase inhibitors in the lysis buffer to preserve phosphorylation signals, and optimize blocking and washing conditions for phospho-specific antibodies.
  • For detection of eNOS phosphorylation at Ser1177, stimulation of endothelial cells with VEGF or exposure to shear stress may be employed; however, the response should be validated in the relevant sample system.
  • In ELISA, ensure that the coating antigen or capture antibody is appropriately selected, and perform a titration to determine optimal working concentrations.
  • As eNOS shares homology with other NOS isoforms, cross-reactivity should be assessed if using in systems with mixed isoform expression.
