Biological Background: Epidermal Growth Factor Receptor Function and Localization
- Epidermal growth factor receptor (EGFR), also known as ErbB-1 or HER1, is a transmembrane receptor tyrosine kinase. An alternatively spliced isoform 2 may act as an antagonist of EGF action.
- Upon binding EGF family ligands (e.g., EGF, TGF-alpha, AREG, BTC, EREG, HBEGF), EGFR triggers homo- or heterodimerization and autophosphorylation, initiating downstream signaling. Related references: PMID:12297049 PMID:15611079 PMID:17909029
- The phosphorylated receptor activates at least four major downstream signaling modules: RAS-RAF-MEK-ERK, PI3K-AKT, PLCgamma-PKC, and STATs, as well as NF-kappa-B. Related references: PMID:27153536 PMID:11116146
- EGFR is primarily located at the cell membrane but also found in the endoplasmic reticulum, Golgi apparatus, endosomes, nucleus, and can be secreted.
- Ubiquitously expressed; isoform 2 is also found in ovarian cancers.
- EGFR undergoes multiple post-translational modifications including phosphorylation, ubiquitination, hydroxylation, methylation, and palmitoylation, as reflected by keywords such as 'Phosphoprotein', 'Ubl conjugation', 'Hydroxylation', 'Methylation', 'Lipoprotein', 'Palmitate', 'Glycoprotein'.
- Acts as a receptor for hepatitis C virus (HCV) entry into hepatocytes.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphopeptide corresponding to the Y992 phosphorylation site of human EGFR. This antibody is expected to specifically recognize the phosphorylated form of EGFR at this residue.
- For Western blot (WB), include appropriate phosphatase inhibitors during sample preparation and consider using a dephosphorylated control to confirm phospho-specificity.
- This antibody has been shown to react with human and mouse EGFR by WB and ELISA. Cross-reactivity with other species or post-translational modifications should be validated empirically in the relevant sample system.
- When performing ELISA, optimizing coating conditions and detection systems is recommended to achieve optimal signal-to-noise ratios.
