Biological Background: EGFR Function and Localization
- EGFR (Epidermal Growth Factor Receptor), also designated ErbB-1 or HER1, is a member of the ErbB family of receptor tyrosine kinases. It is widely expressed in epithelial and other tissues. An alternative isoform (isoform 2) may function as an antagonist of EGF action.
- EGFR binds multiple ligands of the EGF family, including EGF, TGF-α, amphiregulin, and epigen, leading to receptor dimerization and autophosphorylation on cytoplasmic tyrosine residues. Related references: PMID:10805725, PMID:27153536, PMID:2790960, PMID:12297049, PMID:15611079, PMID:17909029
- Upon activation, EGFR recruits adaptor proteins and stimulates major signaling cascades such as RAS-RAF-MEK-ERK, PI3K-AKT, PLCγ-PKC, and STAT pathways, governing proliferation, survival, and differentiation. Related references: PMID:27153536
- EGFR additionally activates NF-kappa-B signaling and can directly phosphorylate substrate proteins including RGS16 (G-protein signaling modulator), MUC1 (cell adhesion/Wnt), and CCDC88A/GIV (migration promoter), linking its activity to diverse cellular processes. Related references: PMID:11116146, PMID:11602604, PMID:11483589, PMID:20462955
- Subcellularly, EGFR is located primarily at the cell membrane but also detected in the endoplasmic reticulum, Golgi, nucleus, endosomes, and can be secreted.
- Ubiquitously expressed in human tissues; the isoform 2 is notably expressed in ovarian cancers.
- (Microbial infection) EGFR serves as a host cell receptor for hepatitis C virus (HCV) entry into hepatocytes by facilitating the CD81-CLDN1 receptor complex formation.
Experimental Guidance and Technical Tips
- This antibody’s immunogen is a synthetic phosphopeptide surrounding Y1086 of human EGFR, designed to specifically recognize the phosphorylated form.
- For Western blot, prepare lysates from EGFR-positive human cell lines (e.g., A431 or HeLa) with and without EGF stimulation; a band at ~134 kDa confirms detection.
- For ELISA, use the antibody to capture or detect phospho-EGFR in human cell lysates or tissue extracts; optimize coating and detection conditions based on your assay platform.
- Ensure phosphatase inhibitor cocktails are included during lysis and sample handling to preserve the phosphorylation signal.
- As cross-reactivity is restricted to human EGFR, validate the antibody in your human sample systems.
