Biological Background: EGFR Function and Localization
- EGFR (ErbB-1/HER1) is a member of the ErbB receptor tyrosine kinase family that mediates cell proliferation, survival, and differentiation upon ligand binding.
- Ligand-induced receptor dimerization triggers autophosphorylation on cytoplasmic tyrosine residues, including Y1068, creating docking sites for adaptor proteins such as GRB2, which activate downstream cascades including RAS-RAF-MEK-ERK, PI3K-AKT, PLCgamma-PKC, and STAT modules. - For immunoprecipitation (IP) and ELISA, validate phospho-specificity using appropriate controls; non-phosphorylated peptide blocking or phosphatase treatment can help differentiate signals.
- Cross-reactivity with mouse ensures applicability in mouse models; validate in the relevant sample system before extensive use.
