Biological Background: EGFR Function and Localization

  • EGFR (ErbB-1/HER1) is a member of the ErbB receptor tyrosine kinase family that mediates cell proliferation, survival, and differentiation upon ligand binding.
  • Ligand-induced receptor dimerization triggers autophosphorylation on cytoplasmic tyrosine residues, including Y1068, creating docking sites for adaptor proteins such as GRB2, which activate downstream cascades including RAS-RAF-MEK-ERK, PI3K-AKT, PLCgamma-PKC, and STAT modules. Related references: PMID:27153536
  • EGFR also signals via NF-kappa-B pathway and directly phosphorylates RGS16 and MUC1, modulating GTPase activity and cell adhesion. Related references: PMID:11116146, PMID:11602604, PMID:11483589
  • Isoform 2 may act as an antagonist of EGF action.
  • Subcellular localization includes plasma membrane, endosomes, endoplasmic reticulum, Golgi, and nucleus, with a secreted form also reported.
  • EGFR is ubiquitously expressed, with isoform 2 notably found in ovarian cancers.
  • Post-translational modifications include phosphorylation, glycosylation, methylation, hydroxylation, palmitoylation, and ubiquitination, typical of a highly regulated membrane receptor.
  • Besides normal physiology, EGFR serves as a host cell receptor for hepatitis C virus (HCV) entry.

Experimental Guidance and Technical Tips

  • For western blotting (WB), use phosphatase inhibitor cocktails during cell lysis to preserve phosphorylation. A specific band near 134 kDa is expected. Confirm specificity by pre-incubating the antibody with the phosphorylated immunogen peptide or by treating lysates with lambda phosphatase.
  • For immunoprecipitation (IP) and ELISA, validate phospho-specificity using appropriate controls; non-phosphorylated peptide blocking or phosphatase treatment can help differentiate signals.
  • Cross-reactivity with mouse ensures applicability in mouse models; validate in the relevant sample system before extensive use.
