Biological Background: DRP1 Function and Localization
- Dynamin-1-like protein (DNM1L), commonly referred to as DRP1, is a member of the dynamin superfamily of large GTPases and is also known as DLP1, DVLP, and Dymple.
- DRP1 mediates mitochondrial and peroxisomal fission by oligomerizing into membrane-associated tubular structures that wrap around scission sites, constricting and severing membranes through a GTP hydrolysis-dependent mechanism. - The immunogen is a synthetic phosphopeptide corresponding to residues around Ser-616 of human DRP1. The antibody is expected to specifically recognize DRP1 only when phosphorylated at this site.
- For western blotting, consider using phosphatase inhibitors during sample preparation to preserve the S616 phosphorylation signal, and validate the band at the predicted molecular weight of 82 kDa in your experimental system.
- In immunoprecipitation, titrate the antibody amount and use optimized lysis buffers to maintain the phospho-epitope; confirm specificity through phosphatase treatment controls.
