Biological Background: Cyclin D1 Function and Localization

  • Cyclin D1 (CCND1) is a G1/S-specific cyclin, also known as B-cell lymphoma 1 protein (BCL-1) and PRAD1 oncogene.
  • It functions as the regulatory subunit of the cyclin D1-CDK4 complex, which phosphorylates retinoblastoma (RB) family proteins to release E2F transcription factors, thereby promoting G1/S transition. Related references: PMID:1827756, PMID:1833066, PMID:19412162
  • Additionally, Cyclin D1 acts as a substrate for SMAD3, repressing SMAD3 transcriptional activity; forms a ternary complex with CDK4 and CDKN1B for nuclear import; and exerts corepressor activity with INSM1 on promoters independently of cell cycle. Related references: PMID:15241418, PMID:9106657, PMID:16569215, PMID:18417529
  • Subcellularly, Cyclin D1 is found in the nucleus, cytoplasm, and nucleus membrane.
  • Post-translational modifications: Cyclin D1 is a phosphoprotein, with documented isopeptide bond and ubiquitin-like conjugation features.
  • Cyclin D1 is a key cell cycle regulator and proto-oncogene, implicated in cancer through chromosomal rearrangements.

Experimental Guidance and Technical Tips

  • This antibody is raised against a synthetic phosphopeptide corresponding to residues surrounding Thr-286 of human Cyclin D1; consider its suitability for detecting endogenous phosphorylated Cyclin D1 in stimulated or cycling cells.
  • For Western blotting, a major band around 34 kDa is expected; inclusion of phosphatase inhibitors in lysis buffer is recommended to preserve phosphorylation signals.
  • In immunohistochemistry (IHC-P), optimal antigen retrieval and blocking conditions should be optimized; phospho-specificity can be confirmed by phosphatase treatment or phospho-blocking peptide competition.
  • ELISA applications may require titration of antibody and antigen concentrations; validate assay specificity with non-phosphorylated peptide controls.
  • Cross-reactivity is indicated for human, mouse, and rat samples; however, sequence alignment of the immunogen with target species is advised to confirm conservation.
