Biological Background: Serine/threonine-protein kinase Chk2 Function and Localization
- Serine/threonine-protein kinase that mediates DNA damage response by phosphorylating downstream effectors at the consensus sequence [L-X-R-X-X-S/T], regulating cell cycle arrest, DNA repair, and apoptosis (PubMed:37943659).
Related references: PMID:37943659 - Controls cell cycle checkpoints by phosphorylating CDC25A, CDC25B, and CDC25C phosphatases, inhibiting their activity and blocking cell cycle progression. Also phosphorylates NEK6 to enforce G2/M arrest.
- Promotes DNA repair through phosphorylation of BRCA2, enhancing RAD51 recruitment for homologous recombination, and activates FOXM1 to stimulate transcription of repair genes.
- Regulates apoptosis by phosphorylating p53 at Ser-20 to alleviate MDM2 inhibition, leading to p53 stabilization; also phosphorylates MDM4 and PML to further promote apoptotic signaling.
- Exhibits tumor suppressor functions: maintains chromosomal stability via BRCA1 phosphorylation for mitotic spindle assembly and is involved in CCAR2-mediated SIRT1 inhibition (PubMed:25361978).
Related references: PMID:25361978 - Under oxidative stress, promotes autophagy by phosphorylating TRIM32 at Ser-55, leading to ATG7 ubiquitination and enhanced autophagosome assembly (PubMed:37943659).
Related references: PMID:37943659 - Also implicated in host-virus interactions: phosphorylates herpes simplex virus protein ICP0, activating its SUMO-targeted ubiquitin ligase activity.
- Localizes predominantly to the nucleus (including PML bodies and nucleoplasm) and is highly expressed in testis, spleen, colon, and peripheral blood leukocytes.
Experimental Guidance and Technical Tips
- To preserve the phosphorylation signal in western blot, include phosphatase inhibitors during sample preparation and consider inducing DNA damage (e.g., by ionizing radiation or chemotherapeutic drugs) to enhance Ser516 phosphorylation.
- For ELISA applications, validate the antibody with phosphorylated peptide standards and appropriate controls; it may be used as either a capture or detection reagent.
- Cross-reactivity with mouse and rat Chk2 has been reported; validate reactivity in your specific experimental system using relevant tissue lysates or cell line extracts.
- Titrate the antibody concentration empirically to achieve optimal signal-to-noise ratios in your assay setup.
