Biological Background: Phospho-Chk1-S345 Function and Localization
- Serine/threonine-protein kinase Chk1, encoded by the CHEK1 gene, is also known as CHK1 checkpoint homolog, cell cycle checkpoint kinase, and checkpoint kinase-1.
- Required for checkpoint-mediated cell cycle arrest and activation of DNA repair in response to DNA damage or unreplicated DNA. Related references: PMID:11535615 PMID:12399544 PMID:12446774
- Binds to and phosphorylates CDC25A, CDC25B, and CDC25C, creating 14-3-3 binding sites that inhibit their phosphatase activity and promote CDC25A degradation, ultimately blocking cell cycle progression. Related references: PMID:9278511 PMID:12676583 PMID:12676925 PMID:12759351 PMID:19734889 PMID:20090422
- Phosphorylates RAD51 at Thr-309, promoting its release from BRCA2 and enhancing chromatin association for homologous recombination repair. Related references: PMID:15665856
- Phosphorylates multiple C-terminal sites of TP53, facilitating its acetylation-dependent activation to induce cell cycle arrest. Related references: PMID:10673501 PMID:15659650 PMID:16511572
- Also phosphorylates NEK6, TLK1, FANCE, SPRTN, and PABIR1/FAM122A, contributing to diverse processes such as chromatin assembly, DNA cross-link repair, and replication stress response. Related references: PMID:18728393 PMID:12660173 PMID:12955071 PMID:17296736 PMID:31316063 PMID:33108758
- Localizes predominantly to the nucleus and chromosome, but is also found in the cytoplasm, cytoskeleton, and centrosome.
- Ubiquitously expressed, with highest levels in thymus, testis, small intestine, and colon.
Experimental Guidance and Technical Tips
- The immunogen design uses a synthetic phosphorylated peptide around Ser345 of human Chk1; confirm specificity by treating lysates with lambda phosphatase to abolish signal.
- For western blot, consider using DNA-damage-inducing agents (e.g., etoposide, hydroxyurea) to enrich phosphorylated Chk1-S345 in human or mouse cell lines.
- ELISA can be performed with captured peptide or lysates; include unphosphorylated peptide as negative control to verify phospho-specificity.
- Species cross-reactivity is reported for human and mouse; validate performance in the target species before large-scale studies.
