Biological Background: Chk1 Function and Localization
- Chk1 is a serine/threonine-protein kinase essential for DNA damage checkpoint-mediated cell cycle arrest and activation of DNA repair. Related references: PMID:11535615, PMID:12399544, PMID:12446774
- It phosphorylates CDC25 phosphatases (CDC25A, CDC25B, CDC25C) on specific serine and threonine residues, creating 14-3-3 binding sites or promoting proteolysis, thereby inhibiting CDK-cyclin complexes and blocking cell cycle progression. Related references: PMID:12676583, PMID:12676925, PMID:12759351, PMID:9278511
- Chk1 also phosphorylates RAD51 at Thr-309, enhancing chromatin association and homologous recombination repair. Related references: PMID:15665856
- It phosphorylates TP53 at multiple C-terminal sites, promoting acetylation and activation of p53-dependent cell cycle arrest and suppression of proliferation. Related references: PMID:10673501, PMID:15659650, PMID:16511572
- Chk1 localizes primarily to the nucleus, chromosomes, cytoplasm, and centrosomes, reflecting its diverse roles in genome maintenance.
- It is ubiquitously expressed with highest abundance in thymus, testis, small intestine, and colon.
- Post-translational modifications include phosphorylation at Ser-280 (autophosphorylation site targeted by this antibody), as well as ubiquitination and acetylation.
- Keywords: ATP-binding, cell cycle, DNA damage, DNA repair, kinase, phosphoprotein, alternative splicing, disease variant, isopeptide bond.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphorylated peptide around Ser280 of human Chk1; consider verifying phospho-specificity using phosphatase-treated samples.
- For WB, include phosphatase inhibitors in the lysis buffer to preserve phosphorylation at Ser280.
- The antibody is specified for WB, IHC-P, and ELISA; optimize dilutions empirically in the relevant sample system.
- Cross-reactivity is reported for human, mouse, and rat; validate in tissues or cell lines known to express Chk1, especially those with high expression (e.g., thymus, testis, small intestine, colon).
