Biological Background: Casein Kinase 2 alpha Function and Localization

  • Casein kinase II subunit alpha (CK2α) is encoded by the CSNK2A1 gene, and is also known as CKII, Cka1, Cka2, CK2A1, or OCNDS. It is a 45 kDa protein that belongs to the serine/threonine kinase family.
  • CK2α is the catalytic subunit of a constitutively active serine/threonine-protein kinase complex that phosphorylates a large number of substrates with acidic residues C-terminal to the phosphoacceptor site, regulating cell cycle progression, apoptosis, transcription, and viral infection. Related references: PMID:11239457, PMID:11704824, PMID:16193064, PMID:12631575, PMID:19387551, PMID:19387552
  • During mitosis, CK2α is required for the p53/TP53-dependent spindle assembly checkpoint that maintains cyclin-B-CDK1 activity and G2 arrest in response to spindle damage. Related references: PMID:11704824, PMID:19188443
  • CK2α regulates apoptosis by phosphorylating p53/TP53 at Ser-392 following UV irradiation and by phosphorylating caspases (CASP9, CASP2) and the apoptotic regulator NOL3, protecting CASP9 from cleavage and inhibiting CASP2 dimerization. Related references: PMID:11239457, PMID:16193064, PMID:22184066
  • It modulates transcription by directly phosphorylating RNA polymerases I, II, III, and IV, as well as numerous transcription factors including NF-kappa-B, STAT1, CREB1, IRF1, IRF2, ATF1, ATF4, SRF, MAX, JUN, FOS, MYC, and MYB. Related references: PMID:12631575, PMID:19387550, PMID:19387551
  • CK2α participates in DNA damage response by phosphorylating DNA repair proteins such as MDC1, MRE11, RAD9A, RAD51, and HTATSF1, facilitating their recruitment to damage sites. Related references: PMID:18411307, PMID:18583988, PMID:18678890
  • CK2α may phosphorylate histone H2A at Ser-1, linking it to chromatin modification. Related references: PMID:38334665
  • CK2α is predominantly localized to the nucleus, and its expression is detected in gastric carcinoma tissue with levels increasing with cancer progression.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic phosphopeptide corresponding to residues surrounding T360 and S362 of human CSNK2A1, ensuring specificity for the dual-phosphorylated form.
  • For Western blot (WB), we recommend using appropriate positive and negative controls (e.g., phosphatase-treated lysates) to confirm phospho-specificity. Optimize antibody dilution in your own experimental system.
  • For ELISA, the antibody can be used to detect phospho-CK2α in cell or tissue lysates; consider pairing with a total CK2α antibody for normalization.
  • Cross-reactivity is reported for human and mouse; validate in other species if needed.
  • Note that the predicted molecular weight is 45 kDa; however, migration may vary depending on gel conditions and phosphorylation state.
