Biological Background: c-Myc Function and Localization

  • Myc proto-oncogene protein (c-Myc), also known as transcription factor p64 or class E basic helix-loop-helix protein 39 (bHLHe39), is a member of the MYC family of transcription factors.
  • Functions as a transcription factor that binds the core DNA sequence 5'-CAC[GA]TG-3' and activates the transcription of growth-related genes, including VEGFA, to promote angiogenesis.
    Related references: PMID:24940000 PMID:25956029
  • Regulates somatic reprogramming and controls self-renewal of embryonic stem cells.
  • Positively regulates alternative splicing of pyruvate kinase PKM by activating HNRNPA1, HNRNPA2, and PTBP1 transcription, thereby inhibiting exon 9 inclusion and promoting the PKM M2 isoform.
    Related references: PMID:20010808
  • Localizes primarily to the nucleus, including the nucleoplasm and nucleolus, but can also be found in the cytoplasm and associated with chromosomes.
  • Subject to multiple post-translational modifications, including phosphorylation at residues such as S62, acetylation, glycosylation, and ubiquitination, which regulate protein stability and transcriptional activity.

Experimental Guidance and Technical Tips

  • The antibody was raised against a synthetic phosphorylated peptide around S62 of human c-Myc, designed for phospho-specificity. For Western blotting, consider using phosphatase inhibitors to preserve phosphorylation status in cell or tissue lysates.
  • Validate the antibody in your sample system with appropriate controls, such as phosphatase-treated samples or non-phosphorylated peptide competition, to confirm phospho-specific detection.
  • For ELISA applications, test antibody binding to phosphorylated versus non-phosphorylated peptide antigens to assess specificity.
