Biological Background: c-Jun Function and Localization
- c-Jun is a core component of the AP-1 transcription factor complex and belongs to the Jun protein family. It recognizes and binds the consensus motif 5'-TGA[GC]TCA-3' to regulate target gene expression. Related references: PMID:10995748 PMID:22083952
- Heterodimerization with FOS family proteins enhances DNA binding and transcriptional activation. Together with FOSB, c-Jun induces FASLG/CD95L expression, promoting activation-induced T cell death. Related references: PMID:12618758
- When phosphorylated at specific residues, c-Jun can modulate downstream signaling. For instance, phosphorylation by HIPK3 promotes NR5A1 activity and steroidogenic gene expression. Related references: PMID:17210646
- c-Jun participates in oncogenic signaling: in colorectal cancer, activated KRAS-mediated transcriptional activation of USP28 involves c-Jun binding to the USP28 promoter. Related references: PMID:24623306
- Subcellularly, c-Jun localizes to the nucleus where it exerts its transcription factor functions.
- c-Jun undergoes post-translational modifications including phosphorylation, acetylation, and ubiquitination, which regulate its activity, stability, and interactions.
- Expressed in developing and adult prostate as well as prostate cancer cells, c-Jun is a proto-oncogene with broad roles in proliferation, differentiation, and apoptosis.
- During Epstein-Barr virus (EBV) infection, c-Jun binds to the viral BZLF1 Z promoter, activating viral gene expression.
Experimental Guidance and Technical Tips
- This antibody was raised against a synthetic peptide phosphorylated at Ser243 of human c-Jun. For Western blot, use appropriate phosphatase inhibitors during sample preparation to preserve phosphorylation.
- Endogenous c-Jun protein weight is approximately 36 kDa; verify band size with the predicted molecular weight and consider potential shifted bands due to phosphorylation or other modifications.
- For IHC-P, antigen retrieval and blocking steps should be optimized. A nuclear staining pattern is expected.
- For ELISA, validate the antibody with both phosphorylated and non-phosphorylated peptide controls to confirm specificity.
- Cross-reactivity with mouse and rat c-Jun is predicted based on sequence homology, but it is advisable to validate in the relevant species and tissue models.
