Biological Background: Tyrosine-protein kinase ABL1 Function and Localization
- Non-receptor tyrosine-protein kinase ABL1, also known as c-Abl, p150, and Abelson tyrosine-protein kinase 1, is encoded by the ABL1 gene. It functions as a key regulator of cytoskeleton dynamics, cell adhesion, motility, endocytosis, autophagy, DNA damage response, and apoptosis.
- ABL1 phosphorylates a wide array of substrates to coordinate actin remodeling. This includes proteins like WASF3 (lamellipodia formation), ANXA1, DBN1, DBNL, CTTN, RAPH1, ENAH, MAPT, and PXN.
- Regulates cell adhesion and motility through phosphorylation of BCAR1, CRK, CRKL, DOK1, EFS, and NEDD9.
Related references:
PMID:22810897 - In response to oxidative stress, ABL1 translocates to mitochondria, leading to mitochondrial dysfunction and cell death. It phosphorylates PRKD2 at Tyr-717.

PMID:28428613 - Translocates to the nucleus where it participates in DNA damage response and apoptosis. It phosphorylates DNA repair proteins (DDB1, DDB2, ERCC3, ERCC6, RAD9A, RAD51, RAD52, WRN) and proapoptotic factor TP73.
- The protein is localized in the cytoplasm, cytoskeleton, nucleus, mitochondrion, and nucleus membrane. It is widely expressed across tissues.
- ABL1 is a proto-oncogene. Chromosomal rearrangement with BCR generates the BCR-ABL fusion protein, a hallmark of chronic myeloid leukemia, leading to constitutive kinase activity.

Experimental Guidance and Technical Tips
- The antibody was raised against a synthetic phosphopeptide corresponding to the region around Y412 of human ABL1. It is predicted to specifically detect phosphorylation at tyrosine 412 in human, mouse, and rat samples.
- For Western blot (WB), immunohistochemistry (IHC-P), and ELISA applications, optimal antibody dilutions and incubation conditions should be determined empirically using positive and negative controls.
- To preserve phosphorylation signals, it is recommended to include phosphatase inhibitors in lysis buffers and to process samples quickly.
- Validate detection in cell lines or tissue samples known to express phospho-c-Abl, such as those stimulated with growth factors or subjected to oxidative stress.
