Biological Background: BRD4 Function and Localization
- BRD4 (Bromodomain-containing protein 4, also known as HUNK1) is a chromatin reader protein that recognizes and binds acetylated histones, playing a key role in epigenetic memory and transcription regulation. Related references: PMID:20871596, PMID:23086925, PMID:23317504
- It remains associated with acetylated chromatin throughout the cell cycle, providing epigenetic memory for postmitotic G1 gene transcription by preserving chromatin structure. Related references: PMID:22334664, PMID:23317504, PMID:23589332
- During interphase, BRD4 regulates transcription of signal-inducible genes by recruiting the P-TEFb complex to promoters and distal enhancers, often in collaboration with JMJD6, displacing negative regulators like HEXIM1 and 7SKsnRNA. Related references: PMID:16109376, PMID:16109377, PMID:19596240
- BRD4 also binds acetylated RELA, activating NF-kappa-B signaling by recruiting P-TEFb. Related references: PMID:19103749
- Following phosphorylation by CK2, BRD4 is recruited to p53/TP53 target promoters, regulating p53-mediated transcription. Related references: PMID:23317504
- In DNA damage response, BRD4 acts as a chromatin insulator by recruiting the condensin-2 complex, thereby inhibiting DNA damage signaling and limiting H2AX phosphorylation spreading.
- Ubiquitously expressed, BRD4 localizes to the nucleus and chromosomes, and undergoes various post-translational modifications including phosphorylation, acetylation, and ubiquitination.
Experimental Guidance and Technical Tips
- For Western blot (WB), use a standard SDS-PAGE protocol with 8% or gradient gels to resolve the ~152 kDa BRD4 protein effectively. Include phosphatase inhibitors in lysis buffers to preserve phosphorylation at T204.
- Validate antibody specificity by testing non-phosphorylated peptide competition or phosphatase treatment.
- For ELISA, consider using phosphorylated and non-phosphorylated peptides to assess specificity; optimize coating concentration.
