Biological Background: Beclin-1 Function and Localization
- Beclin-1, encoded by the BECN1 gene, is also known as Coiled-coil myosin-like BCL2-interacting protein and Protein GT197.
- Central autophagy regulator: acts as a core subunit of the PI3K complex that produces phosphatidylinositol 3-phosphate; PI3KC3-C1 drives autophagosome initiation, while PI3KC3-C2 handles autophagosome maturation and endocytosis. Related references: PMID:18570871, PMID:21358617, PMID:23184933
- Regulates degradative endocytic trafficking and is required for the abscission step in cytokinesis, likely through the PI3KC3-C2 complex. Related references: PMID:20208530, PMID:20643123, PMID:23974797
- Involved in endocytosis. Related references: PMID:25275521
- A 35 kDa mitochondrial fragment (Beclin-1-C) promotes apoptosis by inducing BAX translocation and the release of proapoptotic factors.
- Protects against infection by a neurovirulent strain of Sindbis virus.
- Localizes to multiple compartments: cytoplasm, Golgi apparatus, endosomes, endoplasmic reticulum, mitochondria, autophagosomes, and nucleus.
- Ubiquitously expressed across tissues.
- Undergoes multiple post‑translational modifications including phosphorylation and acetylation; annotated under processes such as autophagy, apoptosis, antiviral defense, cell cycle, and host‑virus interactions.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphopeptide corresponding to Ser93; use phosphatase inhibitors during sample preparation to preserve the phosphorylation signal.
- In WB, the full‑length Beclin-1 band is expected at approximately 52 kDa. Cleavage fragments (e.g., the ~35 kDa mitochondrial form) may also be detected.
- The antibody is validated for WB and ELISA; performance in other applications should be experimentally optimized.
- Cross‑reactivity is confirmed for human; for other species, validate specificity in the relevant sample system.
