Biological Background: Beclin-1 Function and Localization
- Beclin-1 (ATG6/VPS30) is a core component of the class III phosphatidylinositol 3-kinase (PI3KC3) complexes, essential for autophagy and membrane trafficking. Related references: PMID:18570871 PMID:21358617 PMID:23184933
- It regulates distinct steps of autophagy: the PI3KC3-C1 complex initiates autophagosome formation, while PI3KC3-C2 promotes autophagosome maturation and endocytosis. Related references: PMID:20208530 PMID:20643123 PMID:23974797
- Beclin-1 participates in endocytic trafficking and is required for the abscission step of cytokinesis. Related references: PMID:25275521
- A C-terminal 35 kDa fragment of Beclin-1 localizes to mitochondria, promoting apoptosis by facilitating BAX translocation and cytochrome c release.
- Subcellularly, Beclin-1 is found in the cytoplasm, Golgi apparatus, trans-Golgi network, endosomes, endoplasmic reticulum, mitochondria, autophagosomes, and nucleus, reflecting its multifunctional roles.
- Beclin-1 exhibits broad tissue expression (ubiquitous) and is regulated by various post-translational modifications, including phosphorylation (e.g., at Ser90) and acetylation.
- It plays a role in antiviral host defense and protects against neurotropic Sindbis virus infection.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phospho-peptide corresponding to Ser90 of human Beclin-1; therefore, the antibody is expected to specifically recognize the phosphorylated form. Include phosphatase inhibitors in lysis buffers to preserve phosphorylation.
- For Western blot, the predicted molecular weight of full-length Beclin-1 is ~52 kDa. A cleaved ~35 kDa fragment may be detected under apoptotic conditions.
- For ELISA, phosphorylation-specific detection may be confirmed by peptide competition assay using non-phosphorylated and phosphorylated peptides.
- Cross-reactivity with mouse and rat indicates epitope conservation; however, validation in the specific experimental system is recommended.
