Biological Background: ATF-2 Function and Localization
- ATF-2 (Cyclic AMP-dependent transcription factor ATF-2) is a member of the ATF/CREB family of transcription factors; also known as CREB-2, HB16, CRE-BP1, and TREB7.
- It functions as a transcriptional activator, binding to CRE (5'-TGACGTCA-3') or AP-1 (5'-TGACTCA-3') consensus sequences depending on its binding partner, regulating genes involved in anti-apoptosis, cell growth, and DNA damage response.
- In the nucleus, ATF-2 participates in global transcription and DNA damage response. Upon phosphorylation by ATM, it contributes to ionizing radiation-induced S phase checkpoint control and recruitment of the MRN complex to DNA damage foci.
- ATF-2 also localizes to the cytoplasm and mitochondrial outer membrane, where it interacts with HK1 and VDAC1 to impair mitochondrial membrane potential, induce leakage, and promote cell death.
