Biological Background: Amyloid-beta precursor protein Function and Localization
- Amyloid-beta precursor protein (APP) is a type I transmembrane glycoprotein that exists in several isoforms, with APP695 predominating in neurons and APP751/APP770 in non-neuronal cells. It is also known as ABPP, APPI, and cerebral vascular amyloid peptide.
- APP functions as a cell surface receptor involved in neurite growth, neuronal adhesion, and axonogenesis. Interaction between APP molecules on neighboring cells promotes synaptogenesis. Related references: PMID:25122912
- APP acts as a kinesin I membrane receptor, mediating axonal anterograde transport of beta-secretase and presenilin 1, and plays a role in the transport of cargo towards synapses. Related references: PMID:17062754 PMID:23011729
- APP undergoes proteolytic processing to generate amyloid-beta peptides, which are lipophilic metal chelators that bind copper, zinc, and iron. These peptides promote tau aggregation and phosphorylation and can induce oxidative stress and neurotoxicity.
- Subcellularly, APP is localized to the cell membrane, perikaryon, growth cones, clathrin-coated pits, early endosomes, and is also secreted. It can be found in the cell surface, nucleus, and cytoplasm under certain conditions.
- Tissue specificity: APP is highly expressed in the brain and cerebrospinal fluid, with highest levels in the frontal cortex. It is also expressed in kidney, heart, and spleen. Isoform APP695 is neuronal, while APP751 is abundant in T-lymphocytes.
- Phosphorylation at Thr668 (T668) is a key regulatory modification of APP. This antibody specifically recognizes the phosphorylated form, enabling study of phosphorylation-dependent signaling and processing.
Experimental Guidance and Technical Tips
- This antibody was raised against a synthetic phosphopeptide corresponding to residues surrounding Thr668 of human APP. It is suitable for detecting endogenous levels of APP only when phosphorylated at T668.
- For Western blot, a starting dilution range of 1:500–1:2000 is typical for polyclonal antibodies; optimize based on your sample and detection system. Validate specificity by treating cell lysates with lambda phosphatase.
- The predicted molecular weight of full-length APP is ~87 kDa, but post-translational modifications and proteolytic processing can yield multiple bands. Include appropriate molecular weight markers.
- For ELISA, use as capture or detection antibody according to your assay format; pairing with another APP antibody may be necessary for sandwich ELISA.
- Cross-reactivity with mouse and rat has been reported; verify in your target tissue to confirm cross-reactivity and specificity.
- Consider using fresh or properly stored samples, and avoid repeated freeze-thaw cycles to preserve phosphorylation status.
