Biological Background: AKT1 Function and Localization

- For Western blot, a band around 48–55 kDa is expected depending on the isoform and post-translational modifications; consider using phosphatase inhibitors during sample preparation to preserve phosphorylations.
- Validate antibody specificity by treating samples with lambda phosphatase or using non-phosphorylated peptide blocking.
- For ELISA, the antibody can be used to detect phosphorylated AKT in cell lysates; titrate antibody and use appropriate phospho- and total protein controls.
- Cross-reactivity is indicated for human and mouse; additional species may need empirical validation.
