Biological Background: AKT1 Function and Localization
- AKT1, also known as Protein Kinase B alpha (PKBα) or RAC-PK-alpha, is a member of the AKT kinase family comprising three closely related isoforms (AKT1, AKT2, AKT3) that play central roles in regulating metabolism, cell proliferation, survival, growth, and angiogenesis (PubMed:11882383, PubMed:15526160, PubMed:21432781).
Related references: PMID:11882383, PMID:15526160, PMID:21432781 - AKT1 phosphorylates downstream substrates to mediate insulin-induced glucose uptake by promoting SLC2A4/GLUT4 translocation, and regulates glycogen synthesis via inhibition of GSK3A/B (PubMed:11994271).
Related references: PMID:11994271 - It promotes cell survival by phosphorylating and inhibiting pro-apoptotic proteins such as MAP3K5/ASK1 (PubMed:11154276) and BAD (PubMed:10926925), and by enhancing CREB1-dependent transcription of anti-apoptotic genes like BCL2 and MCL1 (PubMed:9829964).
Related references: PMID:11154276, PMID:10926925, PMID:9829964 - AKT1 activates the mTORC1 signaling pathway by phosphorylating TSC2, CASTOR1, and DEPDC5, thereby regulating protein synthesis and cell growth (PubMed:12150915, PubMed:12172553, PubMed:31548394, PubMed:33594058).
Related references: PMID:12150915, PMID:12172553, PMID:31548394, PMID:33594058 - AKT1 phosphorylates FOXO transcription factors, leading to their cytoplasmic retention via 14-3-3 binding, thus inhibiting their pro-apoptotic and cell cycle arrest functions (PubMed:10358075).
Related references: PMID:10358075 - Subcellularly, AKT1 is found in the cytoplasm, nucleus, cell membrane, and mitochondrial intermembrane space, consistent with its diverse signaling roles.
- AKT1 is widely expressed in human tissues and is often overexpressed or hyperactivated in cancers, including prostate and breast cancer, where its phosphorylation at specific residues correlates with disease progression.
Experimental Guidance and Technical Tips
- For Western blotting, prepare lysates with a phosphatase inhibitor cocktail to preserve the phosphorylation at Ser129, and consider using a phospho-specific lysis buffer.
- Validate specificity by comparing signals between phosphatase-treated vs. untreated samples, or by using a non-phosphorylated peptide blocking control.
- In ELISA, ensure the capture or detection format optimally exposes the S129 phosphorylation site; consider pairing with a total AKT1 antibody for normalization.
- The antibody is raised against a synthetic peptide from human AKT1 and is predicted to react with human samples; cross-reactivity with other species should be experimentally verified.
