Biological Background: ACC1 Function and Localization
- Acetyl-CoA carboxylase 1 (ACC1), encoded by the ACACA gene, is a cytosolic enzyme that catalyzes the carboxylation of acetyl-CoA to malonyl-CoA, the first and rate-limiting step of de novo fatty acid biosynthesis. This two-step reaction involves the ATP-dependent carboxylation of the biotin cofactor on the biotin carboxyl carrier (BCC) domain, followed by transfer of the carboxyl group to acetyl-CoA. - ACC1 is a multifunctional enzyme with multiple domains, including biotin carboxylase, biotin carboxyl carrier, and carboxyltransferase, and requires magnesium or manganese as cofactors.
- The enzyme is subject to multiple post-translational modifications, including acetylation and phosphorylation, which modulate its activity and stability.
- ACC1 localizes to the cytoplasm/cytosol.
- Tissue expression includes brain, placenta, skeletal muscle, renal, pancreatic, and adipose tissues; it is expressed at low levels in the lung and not detected in the liver.
- Alternative promoter usage generates different isoforms; the enzyme is allosterically regulated by citrate and palmitoyl-CoA.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphorylated peptide around Ser79; specificity should be confirmed by competition with non-phosphorylated peptide or phosphatase treatment in WB.
- For WB, a 266 kDa band is expected; consider using gradient gels and extended transfer times due to the high molecular weight.
- For ELISA, the antibody may be used for detection of phosphorylated ACC1 in cell lysates; validate in the relevant sample system.
- Cross-reactivity with human, mouse, and rat ACC1 is reported; verify target sequence homology for other species.
