Biological Background: PHD3 Function and Localization

  • Encoded by the EGLN3 gene, PHD3 (also designated HIFPH3/HIFP4H3) is an iron/2-oxoglutarate-dependent dioxygenase belonging to the EGLN family. It contains a prolyl hydroxylase domain and undergoes ubiquitination.
  • Functions as a cellular oxygen sensor: under normoxia, hydroxylates specific proline residues in the oxygen-dependent degradation (ODD) domains of HIF1A and HIF2A, targeting them for VHL-mediated ubiquitination and proteasomal degradation. Under hypoxia, hydroxylation is attenuated, allowing HIFs to stabilize, translocate to the nucleus, and drive hypoxic gene expression. Prefers the CODD site.
