Biological Background: PFKFB3 Function and Localization
- PFKFB3 (6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase 3) is a bifunctional enzyme that catalyzes both the synthesis and degradation of fructose-2,6-bisphosphate, a potent allosteric regulator of glycolysis.
- It is also known as renal carcinoma antigen NY-REN-56 and inducible phosphofructokinase-2 (iPFK-2).
- PFKFB3 belongs to the phosphofructokinase-2/fructose-2,6-bisphosphatase family, which includes four isozymes (PFKFB1-4) with tissue-specific expression; PFKFB3 is ubiquitously expressed.
- By controlling the intracellular concentration of fructose-2,6-bisphosphate, PFKFB3 plays a critical role in regulating glycolytic flux and energy metabolism.
- The enzyme is subject to alternative splicing, generating multiple isoforms (UniProt keyword: Alternative splicing).
- PFKFB3 possesses both kinase (transferase) and bisphosphatase (hydrolase) activities, classifying it as a multifunctional enzyme.
- Regulation occurs through phosphorylation (UniProt keyword: Phosphoprotein); additional keywords include 3D-structure, ATP-binding, Nucleotide-binding, and Reference proteome.
Experimental Guidance and Technical Tips
- The immunogen amino acids 400-500 lie in the C-terminal region, which may be conserved across species, supporting the reported cross-reactivity with human and mouse.
- For WB, expect a specific band at approximately 60 kDa; validate in relevant sample systems (human, mouse) using appropriate loading controls.
- For IHC-P, antigen retrieval may be necessary; consider heat-induced retrieval in citrate buffer (pH 6.0) or EDTA buffer (pH 9.0).
- As a monoclonal antibody, it offers high specificity; always include negative and positive controls in experiments.
- ELISA applications require optimization of coating antigen concentration and antibody dilution to achieve optimal signal-to-noise ratio.
