Biological Background: CD45R/B220 Function and Localization

• CD45R/B220, the B cell isoform of receptor-type tyrosine-protein phosphatase C (PTPRC), is also known as leukocyte common antigen (L-CA), Ly-5, or T200; it is encoded by the Ptprc gene in mice.
• It is a transmembrane glycoprotein with tyrosine phosphatase activity, essential for T-cell activation via the antigen receptor and acts as a positive regulator of T-cell coactivation upon binding to DPP4.
• The protein contains two intracellular phosphatase domains: the first domain has catalytic activity, while the second modulates substrate specificity.
• CD45R/B220 localizes to the cell membrane, membrane rafts, and immunological synapses, facilitating signal transduction during immune responses.
• Alternative splicing generates multiple isoforms with molecular weights ranging from approximately 130 to 144 kDa, and the protein is subject to heavy phosphorylation.
