Biological Background: CD44 Function and Localization
- CD44 (also known as Ly-24, Pgp-1, HERMES, ECMR-III, hyaluronate receptor) is a transmembrane glycoprotein expressed on various immune and epithelial cells, existing in multiple isoforms due to alternative splicing.
- Serves as the primary cell-surface receptor for hyaluronan, mediating cell-cell and cell-matrix adhesion, and regulating cell migration and lymphocyte homing.

PMID:25065622 PMID:8343954 - Engages extracellular matrix components, growth factors, and cytokines, and organizes intracellular signaling complexes that activate RhoGTPases (RAC1, RHOA) and phospholipase C, leading to calcium mobilization and actin cytoskeleton reorganization essential for adhesion and migration.
- Upon binding to LGALS9, triggers LCK-ERK-MAPK signaling to promote NK cell activation.
Related references:
PMID:37006235 - Localizes to the cell membrane, particularly enriched in microvilli, and can also be found as a secreted form.
- Expressed in the intestinal epithelium, with broader tissue distribution in hematopoietic and non-hematopoietic cells.
- Undergoes extensive post-translational modifications including glycosylation, phosphorylation, sulfation, and proteoglycan attachment, which modulate its ligand binding and signaling functions.
Experimental Guidance and Technical Tips
- This antibody is conjugated to PerCP/Cyanine5.5, a tandem dye suitable for flow cytometry. Protect from light and avoid repeated freeze-thaw cycles to maintain fluorophore stability.
- For optimal staining, titrate the antibody on your cell population of interest; typical starting concentrations range from 0.5–5 µg per million cells, but individual optimization is recommended.
- Include appropriate isotype controls and single-stain compensation controls, especially when using tandem dyes, to minimize spectral overlap artifacts.
- CD44 is widely expressed on leukocytes and some epithelial cells; validate target expression in your sample to ensure specific staining and consider using viability dyes to exclude dead cells.
- Ensure proper instrument setup by verifying detector voltages and compensation matrices using your experimental samples.
