Biological Background: CD19 Function and Localization
- CD19 is a B-lymphocyte surface antigen that functions as a coreceptor for the B-cell antigen receptor (BCR) complex, lowering the activation threshold for downstream signaling (By similarity).
- It activates phosphatidylinositol 3-kinase and mobilizes intracellular Ca(2+) stores upon BCR engagement, critical for B-cell responses. Related references: PMID:12387743, PMID:20101619, PMID:9382888
- Although not essential for early B-cell differentiation in bone marrow, CD19 is crucial for the differentiation of B-1 cells. Related references: PMID:7542548, PMID:7543183, PMID:9317126
- It is required for normal B-cell differentiation and proliferation in response to antigen challenges, and for the production of high-affinity antibodies and normal serum immunoglobulin levels. Related references: PMID:12387743, PMID:7542548, PMID:9317126
- CD19 is detected on B cells in spleen, bone marrow, thymus, and lymph nodes, as well as on peripheral blood lymphocytes. Related references: PMID:12387743, PMID:20101619, PMID:7542548, PMID:7543183
- The protein localizes to the cell membrane and membrane rafts, consistent with its role in BCR signaling.
- It is a transmembrane glycoprotein featuring immunoglobulin-like domains, disulfide bonds, and multiple phosphorylation sites.
Experimental Guidance and Technical Tips
- For flow cytometry, consider titrating the antibody to determine the optimal concentration for your specific cell type and experimental conditions.
- Ensure proper compensation settings for the PerCP/Cyanine5.5 tandem dye, as it can be prone to photobleaching; use appropriate isotype controls.
- Validate the staining in relevant mouse sample systems, such as splenocytes or peripheral blood lymphocytes.
- Since the immunogen is full-length recombinant protein, the antibody may recognize both extracellular and intracellular epitopes; permeabilization may be required for intracellular staining.
