Biological Background: CD8a Function and Localization
- CD8 alpha chain is a T-cell surface glycoprotein (also known as T8/Leu-2) that serves as a coreceptor for MHC class I molecules.
- In T-cells, CD8A interacts with the TCR and MHC class I, recruiting the Src kinase LCK to trigger intracellular signaling pathways essential for cytotoxic T-lymphocyte (CTL) activation and target cell elimination.
- In natural killer (NK) cells, CD8A homodimers mediate target cell conjugation and lysis, and promote the survival and differentiation of activated lymphocytes into memory CD8 T-cells.
- The protein is localized at the cell membrane and can also be secreted.
- CD8A is typically expressed as a heterodimer with CD8B on thymus-derived T-cells, but CD8A homodimers are found on subsets of NK cells, memory T-cells, intraepithelial lymphocytes, monocytes, and dendritic cells.
- Post-translational modifications include disulfide bond formation, glycosylation, and palmitoylation, which influence its membrane association and function.
- Alternative splicing generates isoforms with distinct structural properties.
Experimental Guidance and Technical Tips
- The immunogen spans the extracellular domain (amino acids 22–182) of human CD8A, suggesting the antibody recognizes an epitope on the surface-exposed portion, suitable for live-cell staining.
- PerCP/Cyanine5.5 conjugation enables direct detection in flow cytometry, minimizing background and simplifying multicolor panel design.
- Given the cross-reactivity with cynomolgus and rhesus monkey, this antibody may be useful for translational studies in non-human primate models; validate in the relevant sample system.
- Titration is recommended to determine optimal staining concentration and to account for possible batch-to-batch variability.
- Include appropriate isotype controls and single-color compensation controls for accurate data interpretation.
