Biological Background: CD45 Function and Localization

  • CD45, also known as leukocyte common antigen (L-CA) or T200, is a receptor-type tyrosine-protein phosphatase C encoded by the PTPRC gene. Multiple isoforms arise from alternative splicing, with molecular weights ranging from approximately 131 to 147 kDa.
  • CD45 is essential for T-cell activation through the antigen receptor, acting as a positive regulator of coactivation upon binding to DPP4. It recruits and dephosphorylates SKAP1 and FYN, and modulates LYN activity (PubMed:35767951). Related references: PMID:35767951
  • It functions as a protein tyrosine phosphatase, with the first PTPase domain exhibiting enzymatic activity and the second domain influencing substrate specificity.
