Biological Background: CD4 Function and Localization
- T-cell surface glycoprotein CD4, also known as T4 or Leu-3, is a member of the immunoglobulin superfamily and a hallmark marker of T-helper cells.
- Serves as a coreceptor for MHC class II molecules: it simultaneously binds the T-cell receptor (TCR) and MHC class II on antigen-presenting cells, recruiting the Src kinase LCK to initiate intracellular signaling pathways that lead to T-cell activation, lymphokine production, motility, and adhesion.
- Essential for thymic development of T-helper cells and triggers the differentiation of monocytes into functional mature macrophages.
- Acts as the primary receptor for human immunodeficiency virus-1 (HIV-1) by binding the viral envelope glycoprotein gp120, facilitating viral entry. Related references: PMID:12089508 PMID:16331979 PMID:2214026
- Also functions as a receptor for Human Herpesvirus 7 (HHV-7). Related references: PMID:7909607
- Down-regulated by HIV-1 Vpu protein to evade host immune response. Related references: PMID:17346169
- Highly expressed on the surface of T-helper cells, and also present on macrophages, dendritic cells, and NK cells.
- Subcellular localization: cell membrane; contains a single-pass transmembrane domain, disulfide bonds, glycosylation sites, and palmitoylation sites.
Experimental Guidance and Technical Tips
- The antibody is supplied as a ready-to-use PerCP/Cyanine5.5 conjugate, eliminating the need for secondary staining in flow cytometry (FC) assays.
- For FC, consider performing a titration experiment to determine the optimal working concentration in your sample system.
- Include appropriate isotype controls and fluorescence-minus-one (FMO) controls to ensure specificity.
- CD4 expression is most prominent on human T-helper cells; validate in peripheral blood mononuclear cells (PBMCs) or relevant cell lines. Cross-reactivity with Cynomolgus monkey allows for translational research.
- For fixation and permeabilization protocols, verify compatibility with PerCP/Cyanine5.5, as certain buffers may affect fluorophore stability.
