Biological Background: CD38 Function and Localization
- CD38, also known as ADPRC1 or cADPR1, is a multifunctional transmembrane glycoprotein belonging to the ADP-ribosyl cyclase family. It possesses both enzymatic and receptor activities.
- Synthesizes cyclic ADP-ribose (cADPR), a second messenger for glucose-induced insulin secretion. Related references: PMID:7961800 PMID:8253715
- Also synthesizes nicotinate-adenine dinucleotide phosphate (NAADP+), a potent Ca2+ mobilizer, from 2'-phospho-cADPR and nicotinic acid or from NADP+ and nicotinic acid. Related references: PMID:16690024
- Exhibits cADPR hydrolase activity, cleaving cADPR to ADPR. Related references: PMID:7961800 PMID:8253715
- Functions as a receptor for CD31 (PECAM-1) on endothelial cells, promoting lymphocyte activation, proliferation, and transendothelial migration. Related references: PMID:9551996
- Involved in dendritic cell maturation, regulating CCL21-driven migration, cell survival, and Th1-polarizing activity. Related references: PMID:16293598
- In lamina propria T lymphocytes, CD38/CD31 interactions trigger a signaling cascade involving LCK and LAT, leading to cytokine release. Related references: PMID:11259373
- Localizes to the cell surface and plasma membrane, with high expression in pancreas, liver, kidney, brain, testis, ovary, placenta, and certain tumors. Undergoes alternative splicing and contains disulfide bonds and glycosylation sites.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (aa 43-300) of human CD38, which contains the catalytic site and receptor-binding regions. This design may enhance reactivity against native cell surface CD38 in flow cytometry.
- For flow cytometry (FC), use this PerCP/Cyanine5.5-conjugated antibody to detect CD38 on live cells without the need for a secondary antibody. Include appropriate isotype controls and titrate the antibody to optimize staining.
- Validate the antibody in the relevant sample system (e.g., human or cynomolgus PBMCs, cell lines expressing CD38) and confirm specificity using blocking peptides or CD38-negative cells.
