CD94 (KLRD1) is a C-type lectin-like transmembrane receptor that forms disulfide-linked heterodimers with NKG2A (KLRC1) or NKG2C (KLRC2). These complexes recognize the non-classical MHC class I molecule HLA-E presenting self-peptides, enabling immune cells to monitor MHC class I expression. The KLRD1-KLRC1 heterodimer serves as an inhibitory receptor. On NK cells, it recruits tyrosine phosphatases via ITIM motifs to dampen activation signals.
