Biological Background: CD25 Function and Localization
- CD25, also known as Interleukin-2 receptor subunit alpha or p55, is encoded by the Il2ra gene. It serves as the alpha chain of the high-affinity IL-2 receptor complex.
- CD25 is a critical regulator of immune tolerance, primarily through its role in controlling the activity of regulatory T cells (TREGs). TREGs suppress the activation and expansion of autoreactive T cells.
- The protein is a single-pass type I transmembrane receptor, localized to the cell membrane, and belongs to the type I cytokine receptor family.
- CD25 contains structural features such as a signal peptide, two sushi (CCP/SCR) domains, and a transmembrane helix. It is subject to post-translational modifications including N-linked glycosylation and disulfide bond formation.
- As a receptor for interleukin-2 (IL-2), CD25 is pivotal in lymphocyte proliferation, differentiation, and survival, making it a key target in immunology research.
- The mouse CD25 protein shares significant homology with human CD25 (encoded by Gene ID 3559), and this antibody specifically recognizes the mouse form.
Experimental Guidance and Technical Tips
- This PerCP/Cyanine5.5-conjugated monoclonal antibody is specifically designed for flow cytometry (FC) applications, enabling direct detection of mouse CD25 on the cell surface without the need for a secondary antibody.
- The immunogen used was a recombinant fusion protein corresponding to the extracellular domain (amino acids 22-236) of mouse CD25, which encompasses the IL-2 binding region and is likely to generate antibodies recognizing the native conformation.
- For flow cytometry, consider titrating the antibody to determine the optimal concentration for your specific cell type and assay conditions. Use appropriate isotype controls and viable cell gating strategies to ensure specificity.
- Note that CD25 is constitutively expressed on regulatory T cells and upregulated on activated T and B cells. Validate detection in your relevant cell system, such as mouse splenocytes or purified T cell populations.
