Biological Background: CD8a Function and Localization

• CD8a (T-cell surface glycoprotein CD8 alpha chain) is a transmembrane glycoprotein encoded by the CD8A gene (Gene ID: 925). It is also known as T8/Leu-2 and p32.
• Serves as a co-receptor for MHC class I molecules on antigen-presenting cells, interacting simultaneously with the T-cell receptor (TCR) to recruit the Src kinase LCK, initiating intracellular signaling that leads to cytotoxic T-lymphocyte activation, cytokine production, motility, and adhesion.
• Plays a critical role in adaptive immunity by enabling cytotoxic T-lymphocytes to recognize and eliminate infected cells and tumor cells.
• In natural killer (NK) cells, CD8A homodimers provide a survival mechanism that allows conjugation and lysis of multiple target cells, and promote the survival and differentiation of activated lymphocytes into memory CD8 T-cells.
• CD8 is typically a disulfide-linked heterodimer of alpha (CD8A) and beta (CD8B) chains on thymus-derived T-cells, but can also be expressed as a CD8A homodimer on subsets of NK cells, memory T-cells, intraepithelial lymphocytes, monocytes, and dendritic cells.
• Subcellular localization includes the cell membrane and secreted forms; it is a lipoprotein with a transmembrane helix, and undergoes post-translational modifications such as glycosylation, disulfide bond formation, and palmitoylation.
• Belongs to the immunoglobulin superfamily, containing an immunoglobulin domain. Alternative splicing produces multiple isoforms.
• Expressed on plasmacytoid dendritic cells upon herpes simplex virus-1 stimulation, indicating a role in antiviral responses.

Experimental Guidance and Technical Tips

• The immunogen (amino acids 22-182) covers the extracellular domain of CD8A, which is predicted to generate antibodies recognizing the native cell surface protein. For flow cytometry, consider titrating the antibody to determine the optimal concentration for your specific cell type and staining protocol.
• As this antibody is conjugated to PerCP, ensure that your flow cytometer is equipped with appropriate laser and filter sets (typically 488 nm excitation, 675 nm emission). PerCP is susceptible to photobleaching; handle stained samples in the dark and avoid prolonged exposure to light.
• Cross-reactivity has been validated for human and cynomolgus monkey. If using other species, verify sequence homology of the immunogen region to predict potential cross-reactivity and validate experimentally.
• For intracellular staining, a permeabilization step may be required. Include appropriate isotype controls to distinguish specific staining.
