Biological Background: CD4 Function and Localization

  • T-cell surface glycoprotein CD4 (CD4) is a 51 kDa type I transmembrane glycoprotein and a member of the immunoglobulin superfamily. It is also known as T4, Leu-3, and OKT4D.
  • CD4 serves as a co-receptor for the T-cell receptor (TCR) by binding simultaneously to MHC class II molecules on antigen-presenting cells, thereby enhancing T-cell sensitivity and activation. Upon engagement, it recruits the Src kinase LCK to the TCR-CD3 complex, initiating intracellular signaling cascades that promote lymphokine production, motility, adhesion, and T-helper cell differentiation.
  • CD4 is primarily expressed on the surface of T-helper cells, but can also be found on macrophages, dendritic cells, and natural killer (NK) cells, where it may participate in differentiation, activation, and migration in a TCR-independent manner.
  • In the thymus, CD4 plays an essential role in the positive selection and maturation of T-helper cells, ensuring proper adaptive immune function.
  • As a key host factor in viral pathogenesis, CD4 functions as the primary receptor for human immunodeficiency virus-1 (HIV-1), facilitating viral entry.
