Biological Background: CD31/PECAM1 Function and Localization
- PECAM1 (CD31) is an 81 kDa cell adhesion molecule and member of the immunoglobulin superfamily, with alternative splicing generating multiple isoforms.
- Essential for leukocyte transendothelial migration (TEM) during inflammation; Tyr-679 phosphorylation is critical for trafficking from the lateral border recycling compartment (LBRC) to sites of leukocyte passage.
- Mediates homophilic endothelial cell-cell adhesion at junctions and heterophilic interaction with CD177 to facilitate neutrophil TEM.
- Prevents macrophage-mediated phagocytosis of viable leukocytes through detachment signaling, while promoting phagocytosis of apoptotic cells by tethering them to phagocytes.
- Modulates bradykinin receptor BDKRB2 activation and hyperosmotic stress-induced ERK1/2 signaling in endothelial cells.

- Subcellularly located at cell membrane, membrane rafts, and cell junctions.
