Biological Background: CD95/FAS Function and Localization

  • Tumor necrosis factor receptor superfamily member 6 (FAS, also known as CD95, Apo-1, APT1, TNFR6, Tnfrsf6) is encoded by the Fas gene in mouse.
  • Functions as a receptor for TNFSF6/FASLG; upon ligand binding, recruits FADD and activates caspase CASP8 via the death-inducing signaling complex (DISC), initiating the caspase cascade that leads to apoptosis.
  • FAS-mediated apoptosis may contribute to peripheral tolerance and the antigen-stimulated suicide of mature T-cells.
  • Localizes to the cell membrane and membrane rafts.
  • Detected in various tissues including thymus, liver, lung, heart, and adult ovary.
  • Post-translational modifications include disulfide bonds, glycosylation, and palmitoylation (glycoprotein, lipoprotein, palmitate, phosphoprotein).
  • Belongs to the tumor necrosis factor receptor superfamily; contains extracellular cysteine-rich repeats characteristic of the family.
