- T-cell surface glycoprotein CD4 (also designated L3T4 and Ly-4) is a type I transmembrane protein belonging to the immunoglobulin superfamily.
- Functions as a co-receptor for MHC class II molecules on antigen-presenting cells, enhancing T-cell receptor (TCR) signaling.
- Upon ligand engagement, CD4 recruits the Src kinase LCK to the TCR-CD3 complex, initiating phosphorylation cascades that promote T-cell activation, lymphokine production, and adhesion.
- Plays a critical role in thymic T-helper cell development and triggers monocyte-to-macrophage differentiation in a TCR-independent manner.
- Primarily expressed on the surface of T-helper cells, but also present on macrophages, dendritic cells, and NK cells.
- Subcellular location is the plasma membrane, with a single-pass transmembrane domain and a cytoplasmic tail involved in signal transduction.
