Biological Background: CD150/SLAM Function and Localization
- CD150 (SLAMF1) is a self-ligand receptor of the SLAM family, modulating immune cell activation and differentiation through homo- or heterotypic interactions. Its signaling is regulated by small cytoplasmic adapters SH2D1A/SAP and SH2D1B/EAT-2, leading to distinct outcomes in T- and B-lymphocytes.
- In T-cells, CD150 mediates IL-2-independent proliferation and IFN-gamma production. Downstream signaling involves INPP5D, DOK1, and DOK2 inhibiting IFN-gamma, while PRKCQ, BCL10, and NFKB1 promote T-cell activation and Th2 cytokine production. - Additionally, CD150 promotes T-cell receptor-induced IL-4 secretion by CD4+ cells and is required for IL-4 production by germinal center T follicular helper cells. Related references: PMID:15123745 PMID:20525889
- In B-cells, CD150 activates ERK signaling independently of SH2D1A but involving SYK and INPP5D, and activates Akt signaling dependent on SYK and SH2D1A.
