Biological Background: CD150/SLAM Function and Localization

  • CD150 (SLAMF1) is a self-ligand receptor of the SLAM family, modulating immune cell activation and differentiation through homo- or heterotypic interactions. Its signaling is regulated by small cytoplasmic adapters SH2D1A/SAP and SH2D1B/EAT-2, leading to distinct outcomes in T- and B-lymphocytes.
  • In T-cells, CD150 mediates IL-2-independent proliferation and IFN-gamma production. Downstream signaling involves INPP5D, DOK1, and DOK2 inhibiting IFN-gamma, while PRKCQ, BCL10, and NFKB1 promote T-cell activation and Th2 cytokine production. Related references: PMID:12351401 PMID:9126961 PMID:11477403 PMID:15539155 PMID:16847311
  • Additionally, CD150 promotes T-cell receptor-induced IL-4 secretion by CD4+ cells and is required for IL-4 production by germinal center T follicular helper cells. Related references: PMID:15123745 PMID:20525889
  • In B-cells, CD150 activates ERK signaling independently of SH2D1A but involving SYK and INPP5D, and activates Akt signaling dependent on SYK and SH2D1A. Together with CD84 and SLAMF6, it may act as a negative regulator of the humoral immune response. Related references: PMID:15315965 PMID:25926831
  • In macrophages, CD150 down-regulates IL-12, TNF, and nitric oxide in response to LPS and participates in innate immunity against Gram-negative bacteria by recognizing bacterial OmpC/OmpF, regulating phagosome maturation, and recruiting the PI3K complex II for NOX2 activity. Related references: PMID:15123745 PMID:20818396 PMID:22493499
  • CD150 is localized at the cell membrane and classified as a transmembrane receptor with immunoglobulin domains, disulfide bonds, and glycosylation; it is a phosphoprotein involved in cell adhesion and phagocytosis.
  • It plays a role in allergic responses by regulating allergen-induced Th2 and Th1 cytokine secretion and is involved in the development of natural killer T cells. Related references: PMID:16528012 PMID:18031695 PMID:18606638

Experimental Guidance and Technical Tips

  • Flow cytometry: This PE/Cyanine7-conjugated antibody can be used directly for surface staining of mouse cells expressing CD150. Pre-titration is recommended to determine the optimal concentration for your assay. Ensure proper compensation when combining with other fluorophores.
  • Immunogen design: The immunogen corresponds to the extracellular region (aa 25-242) encompassing immunoglobulin-like domains, suggesting that the recognized epitopes are likely accessible on live cells. Validate staining patterns in the relevant sample system.
  • Species cross-reactivity: This antibody was raised against mouse CD150. Reactivity with other species has not been specified; if used beyond mouse, cross-reactivity should be confirmed experimentally.
