Biological Background: Endoglin Function and Localization

  • Endoglin is a transmembrane glycoprotein primarily expressed on vascular endothelial cells, also known as cell surface MJ7/18 antigen.
  • It plays an essential role in angiogenesis and is required for normal extraembryonic angiogenesis and embryonic heart development. Related references: PMID:10625534
  • Endoglin regulates the migration of vascular endothelial cells, contributing to vascular remodeling. Related references: PMID:17540773
  • It may regulate endothelial cell shape changes in response to blood flow, driving establishment of normal vascular morphology during angiogenesis. Related references: PMID:28530658
  • Acts as a TGF-beta coreceptor, involved in TGF-beta/BMP signaling cascades leading to SMAD transcription factor activation. Related references: PMID:23300529
  • Required for GDF2/BMP9 signaling through SMAD1 and modulates TGFB1 signaling through SMAD3.
  • Subcellularly localized to the cell membrane; detected on endothelial cells of adult pulmonary artery, heart capillaries, and lung alveolar capillaries; also found in stromal cells of various organs. Related references: PMID:8194490, PMID:10625534
  • Post-translational modifications include glycosylation and phosphorylation; classified as a disulfide-linked glycoprotein.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (amino acids 27-581) of mouse CD105, which may facilitate recognition of the native cell surface protein in flow cytometry.
  • For flow cytometry, consider using appropriate isotype controls and titrate the antibody to determine optimal staining concentration for your cell type.
  • Since CD105 is expressed on endothelial cells and some stromal cells, ensure proper gating strategies and include endothelial markers for identification.
  • The antibody is validated for mouse reactivity; cross-reactivity with other species has not been tested. Validate in your experimental system if using other species.
