  • CD4 is an integral membrane glycoprotein essential for adaptive immune responses, acting as a coreceptor for MHC class II molecules on antigen-presenting cells to facilitate T-cell receptor (TCR) signaling.
  • Upon interaction with MHC class II, CD4 recruits the Src kinase LCK to the TCR-CD3 complex, initiating intracellular signaling cascades that promote T-helper cell activation, lymphokine production, motility, and adhesion.
  • In macrophages and NK cells, CD4 plays a TCR/LCK-independent role in differentiation, activation, cytokine expression, and cell migration.
