Biological Background: CD4 Function and Localization
- CD4 is an integral membrane glycoprotein essential for adaptive immune responses, acting as a coreceptor for MHC class II molecules on antigen-presenting cells to facilitate T-cell receptor (TCR) signaling.
- Upon interaction with MHC class II, CD4 recruits the Src kinase LCK to the TCR-CD3 complex, initiating intracellular signaling cascades that promote T-helper cell activation, lymphokine production, motility, and adhesion.
- In macrophages and NK cells, CD4 plays a TCR/LCK-independent role in differentiation, activation, cytokine expression, and cell migration.
- CD4 is the primary receptor for HIV-1, mediating viral entry (PubMed:12089508, PubMed:16331979, PubMed:2214026), and is down-regulated by HIV-1 Vpu (PubMed:17346169). It also serves as a receptor for Human Herpesvirus 7 (PubMed:7909607).
