Biological Background: CD33 Function and Localization

  • CD33 (Siglec-3) is a sialic acid-binding immunoglobulin-like lectin that mediates cell-cell interactions and helps maintain immune cells in a resting state. Related references: PMID:10611343, PMID:11320212, PMID:15597323
  • It preferentially binds alpha-2,3- and alpha-2,6-linked sialic acid-bearing glycans on glycoproteins and glycolipids. Related references: PMID:7718872
  • Upon ligand engagement (e.g., C1q or sialylated glycoproteins), its cytoplasmic ITIM motifs are phosphorylated by Src-like kinases such as LCK, creating docking sites for phosphatases PTPN6/SHP-1 and PTPN11/SHP-2. Related references: PMID:10887109, PMID:28325905, PMID:10206955, PMID:10556798
  • These phosphatases dephosphorylate downstream signaling molecules, leading to inhibitory effects; one repressive pathway in monocytes requires PI3K. Related references: PMID:10206955, PMID:10887109, PMID:15597323
  • CD33 localizes to the cell membrane and peroxisomes, and is expressed primarily on monocytic/myeloid lineage cells and brain microglia (alternative splicing produces multiple isoforms).
  • The protein is a single-pass type I membrane glycoprotein with an extracellular Ig-like V-type domain, several intracellular ITIM motifs, and is modified by disulfide bonds and phosphorylation.

Experimental Guidance and Technical Tips

  • The immunogen covers amino acids 18–259, which includes the extracellular sialic acid-binding domain, so the antibody is likely to recognize native CD33 on the cell surface—ideal for flow cytometry (FC).
  • For optimal FC staining, titrate the antibody in the relevant human myeloid/monocytic cell system to determine the appropriate working dilution.
  • Include proper isotype controls and viability dyes in your FC panel, and protect the PE/Cyanine7-conjugated antibody from light, as the fluorophore is photosensitive.
