Biological Background: AXL Function and Localization

• AXL (also known as UFO, ARK, JTK11, Tyro7) is a receptor tyrosine kinase and proto-oncogene encoded by the AXL gene, belonging to the TAM family (Tyro3, AXL, Mer).
• Functions as a receptor for growth arrest-specific protein 6 (GAS6); ligand binding induces receptor dimerization, autophosphorylation, and activation of multiple downstream pathways including PI3K/AKT, GRB2, PLCG1, LCK, and PTPN11, regulating cell survival, proliferation, migration, and differentiation.
• GAS6/AXL signaling participates in endothelial cell survival under acidification, natural killer cell development, hepatic regeneration, GnRH neuron migration, platelet activation, and negative regulation of Toll-like receptor (TLR)-mediated innate immune responses.
